This chapter examines the nature of interactions between proteins and how proteins function in the cell. Many proteins that control disease processes but are considered undruggable are not enzymes. This means that these proteins do not have an obvious pocket on their surface in which a small molecule can bind. Instead, these proteins function in the cell by interacting with other proteins. Blocking such protein-protein interactions has been considered to be one of the most challenging tasks for drugs. The chapter first traces the origins of the search for inhibitors of protein-protein interactions and proceeds by explaining how individual proteins assemble into larger protein complexes. It then discusses the challenge of inhibiting protein interactions with small molecule drugs, along with the protein interactions of the RAS proteins. The chapter concludes by looking at the discovery of the p53 protein and the MDM2 protein.
